Why this happens

The old USP colour 'heavy-metals' test is retired; regulators now require a Q3D risk assessment with permitted daily exposures (PDEs) per element and route of administration — a frequent deficiency when it is missing or generic.

What to do, step by step

  1. List every potential elemental source: API synthesis catalysts, reagents, water, excipients, manufacturing equipment, and container-closure.
  2. Take each element's PDE for the intended route (oral / parenteral / inhalation) from ICH Q3D Table A.2.2.
  3. Convert the PDE to a concentration limit using the maximum daily dose; focus testing on Class 1 (As, Cd, Hg, Pb) plus any catalyst actually used.
  4. Document the risk assessment and control strategy; only routine-test where the assessment shows a real risk of exceeding 30% of the PDE.
