Do I even need a full BE study — or does a BCS biowaiver / in-vitro comparison apply?

Why this happens

Running a BE study you did not need wastes months and budget; skipping one you did need gets the dossier rejected. The BCS class of the API and the formulation decide which path is defensible.

إجراء دراسة تكافؤ لا لزوم لها يهدر أشهرًا وميزانية، وتخطّي دراسة كنت تحتاجها يؤدي لرفض الملف. وتصنيف BCS للمادة الفعّالة والتركيبة هما ما يحدّد المسار المدافَع عنه.

What to do, step by step

1. Classify the API by BCS (solubility + permeability). Class I and (conditionally) Class III immediate-release products may qualify for a biowaiver.
2. Confirm the product is immediate-release, not a narrow-therapeutic-index drug, and has no absorption-critical excipients.
3. For a biowaiver, generate comparative dissolution across three media (pH 1.2 / 4.5 / 6.8) and demonstrate similarity (f2 ≥ 50 or both ≥ 85% in 15 min).
4. For additional strengths of a proportionally similar formulation, apply the strength-biowaiver to avoid repeating in-vivo studies.

1. صنّف المادة الفعّالة وفق BCS (الذوبانية + النفاذية). قد تتأهل منتجات الإطلاق الفوري من الصنف الأول و(بشروط) الثالث للإعفاء الحيوي.
2. تأكّد أن المنتج فوري الإطلاق، وليس ضيّق النافذة العلاجية، وخالٍ من سواغات حرجة للامتصاص.
3. للإعفاء الحيوي، أنشئ منحنيات تذويب مقارنة في ثلاثة أوساط (pH 1.2 / 4.5 / 6.8) وأثبت التشابه (f2 ≥ 50 أو بلوغ كليهما ≥ 85% خلال 15 دقيقة).
4. للقوى الإضافية من تركيبة متناسبة، طبّق إعفاء القوة لتجنّب تكرار الدراسات داخل الجسم.
