Why this happens

Single-source dependence is the industry's most common supply risk. A supplier GMP failure, a suspended CEP, an impurity/nitrosamine finding, or a price/geopolitical shock can halt your product overnight.

What to do, step by step

  1. Identify alternative manufacturers holding a valid CEP (EDQM database) or a US/EU/relevant DMF for the same API.
  2. Screen candidates on GMP status (EU/PIC/S/WHO), impurity profile, polymorphic form, particle-size distribution and a nitrosamine risk assessment.
  3. Qualify the second source: obtain samples, run comparative characterization and stability, and confirm no impact on the finished-product BE.
  4. File the change of API source as a variation (CTD Module 3.2.S) in each market — use reliance/reference where the first market's approval is accepted.
  5. Bridge supply meanwhile: interim/safety stock, and adopt a dual-sourcing policy so this never recurs.

  5. جسّر التوريد في الأثناء: مخزون احتياطي/آمن، واعتمد سياسة ازدواج المصادر حتى لا يتكرّر ذلك.
